Arbutin is a glycosylated form of hydroquinone found in Arctostaphylos plants, known for its various biological activities, including inhibiting tyrosinase, offering antioxidant properties, and providing anti-inflammatory effects. It effectively inhibits human tyrosinase activity in crude tyrosinase solutions isolated from human melanocytes (IC50s = 5.7 and 18.9 mM for L-tyrosine and L-DOPA substrates, respectively), as well as in intact melanocytes (IC50 = 0.5 mM). Arbutin (50 μM) also inhibits hemolysis induced by the free radical generator AAPH in sheep erythrocytes and protects cultured human skin fibroblasts from AAPH-induced cell viability loss at concentrations above 125 μM. In a rat model of acute lung injury induced by LPS, arbutin (50 mg/kg) reduces the levels of IL-1β, IL-6, and TNF-α in lung tissue and serum. Arbutin-containing formulations are commonly used in the treatment of hyperpigmentation disorders.
